Why does DNA replication occur discontinuously on one strand…

Answer= At the replication fork:

Leading strand

Template is oriented 3 5

DNA polymerase synthesizes DNA continuously toward the fork

Lagging strand

Template is oriented 5 3

DNA polymerase cannot synthesize continuously

So DNA is made in short fragments called Okazaki fragments hence,

eplication is discontinuous

Role of enzymes involved

Helicase

Unwinds the double-stranded DNA

Primase

Synthesizes short RNA primers on the lagging strand

DNA polymerase III

Extends the primers to form Okazaki fragments (5 3)

DNA polymerase I

Removes RNA primers and replaces them with DNA

DNA ligase

Joins the Okazaki fragments into a continuous
